Effects of Some Complex Organic Nitrogenous Substances on Growth of Cymbidium seidenfadenii (P.J.Cribb & Du Puy) P.J.Cribb Protocorms and Seedlings in vitro

Abstract

In vitro protocorms and seedlings of Cymbidium seidenfadenii (P.J.Cribb & Du Puy) P.J.Cribb were cultured on semi-solid MS medium supplemented with 0.5, 1.0 and 2.0 g/L peptone, casein hydrolysate and yeast extract respectively for 12 weeks. The results showed that the highest number of protocorms multiplication could induce when cultured on the medium added with 2.0 g/L casein hydrolysate or 0.5 g/L yeast extract (7.1+1.7 and 7.1+1.0 protocorms, respectively). However, the highest number of shoots and leaves derived from protocorms (2.1 + 0.7 shoots and 4.3 + 1.4 leaves, respectively) were found on the medium without organic substances. In case of seedling explants, the highest number of protocorms (2.1 + 1.0 protocorms) was observed on the augment medium with 1.0 g/L casein hydrolysate. However, the highest leaf and shoot length (0.87 + 0.04 cm and 2.81 + 0.01 cm, respectively) was noticed on the medium supplemented with 2.0 g/L Peptone. Keywords:  Dendrobium seidenfadenii (P.J.Cribb & Du Puy) P.J.Cribb,    complex organic nitrogenous substances, growth
