Effects of Culture Media and Plant Growth Regulators on In Vitro Seedling Growth of Pholidota imbricata Lindl.

Abstract

The effect of different media (MS, ½MS, VW and ½VW) on growth of in vitro seedling culture of Pholidota imbricata Lindl. was studied for 8 weeks. The results showed that the highest average of shoot numbers (1.68 shoots/plant) (47.5%), leaf numbers (5.18 leaves/plant) and root numbers (7.60 roots/plant) (100%) and shoot length (3.81 cm) were found in plantlet culture on the ½VW medium. Moreover, effect of different types and concentrations of cytokinins (BA, kinetin, TDZ) on in vitro seedling culture of P. imbricata Lindl. on ½VW medium for 8 weeks was also investigated. The results showed that the highest percentage of pseudobulb formation (30%), shoot numbers (1.68 shoots/plant) (60%) and leaf numbers (4.25 leaves/plant) were obtained when cultured on the medium with 0.1 mg/L TDZ. The seedling culture of P. imbricata Lindl. on ½VW medium supplemented with different types and concentrations of auxins (NAA, IAA, IBA) for 8 weeks demonstrated that highest average of shoot numbers (1.63 shoots/plant), the percentage of shoot formation (32.5%) and leaf numbers (6.55 leaves/plant) were observed when cultured on the medium with 0.1 mg/L IAA.
